Multicomponent One-pot Reactions Towards the Synthesis of Stereoisomers of Dipicolylamine Complexes. - Chemistry, an Asian journal
Reported are multi-component one-pot syntheses of chiral complexes [M(L(R) OR')Cl2 ] or [M(L(R) SR')Cl2 ] from the mixture of an N-substituted ethylenediamine, pyridine-2-carboxaldehyde, a primary alcohol or thiol and MCl2 utilizing in-situ formed cyclized Schiff bases where a C-O bond, two stereocenters, and three C-N bonds are formed (M=Zn, Cu, Ni, Cd; R=Et, Ph; R'=Me, Et, nPr, nBu). Tridentate ligands L(R) OR' and L(R) SR' comprise two chiral centers and a hemiaminal ether or hemiaminal thioether moiety on the dipicolylamine skeleton. Syn-[Zn(L(Ph) OMe)Cl2 ] precipitates out readily from the reaction mixture as a major product whereas anti-[Zn(L(Ph) OMe)Cl2 ] stays in solution as minor product. Both syn-[Zn(L(Ph) OMe)Cl2 ] and anti-[Zn(L(Ph) OMe)Cl2 ] were characterized using NMR spectroscopy and mass spectrometry. Solid-state structures revealed that syn-[Zn(L(Ph) OMe)Cl2 ] adopted a square pyramidal geometry while anti-[Zn(L(Ph) OMe)Cl2 ] possesses a trigonal bipyramidal geometry around the Zn centers. The scope of this method was shown to be wide by varying the components of the dynamic coordination assembly, and the structures of the complexes isolated were confirmed by NMR spectroscopy, mass spectrometry, and X-ray crystallography. Syn complexes were isolated as major products with Zn(II) and Cu(II) , and anti complexes were found to be major products with Ni(II) and Cd(II) . Hemiaminals and hemiaminal ethers are known to be unstable and are seldom observed as part of cyclic organic compounds or as coordinated ligands assembled around metals. It is now shown, with the support of experimental results, that linear hemiaminal ethers or thioethers can be assembled without the assistance of Lewis acidic metals in the multi-component assembly, and a possible pathway of the formation of hemiaminal ethers has been proposed.© 2016 WILEY-VCH Verlag GmbH & Co. Production, optimization and characterization of fibrinolytic enzyme by Bacillus subtilis RJAS19. - Pakistan journal of biological sciences : PJBS
The present study aimed at the production, purification and characterization of fibrinolytic nattokinase enzyme from the bacteria isolated from natto food. For the purpose, a fibrinolytic bacterium was isolated and identified as Bacillus subtilis based on 16S rDNA sequence analysis. The strain was employed for the production and optimization of fibrinolytic enzyme. The strain showed better enzyme production during 72nd h of incubation time with 50 degrees C at the pH 9. The lactose and peptone were found to be increasing the enzyme production rate. The enzyme produced was purified and also characterized with the help of SDS-PAGE analysis. The activity and stability profile of the purified enzyme was tested against different temperature and pH. The observations suggesting that the potential of fibrinolytic enzyme produced by Bacillus subtilis RJAS 19 for its applications in preventive medicines.
Receiving or not receiving acupuncture in a trial: the experience of participants recovering from breast cancer treatment. - Complementary therapies in clinical practice
To explore the experience of breast cancer patients who either received acupuncture or were allocated to the control group.Focus group/interviews nested within our multi-site randomised controlled trial. Participants were recruited from the standard care and experimental arm. The interviews/focus groups were transcribed and analysed thematically.Of the 302 eligible participants 13% (n = 40) contributed to the focus groups/interviews, across three study sites in the UK. Five common themes were identified, including: drivers to take part, the experience of receiving acupuncture, being allocated standard care (control) and reflections on taking part in the trial. The subgroup of control group participants (n = 9) reported disappointment on hearing their allocation, but recognised the value of their role to the study.Recipients of acupuncture reported beneficial effects in managing fatigue and related symptoms. The finding that control participants were disappointed warrants further investigation and consideration when designing interventional studies.Copyright © 2014 Elsevier Ltd. All rights reserved.
Importance of halide involving interactions at Hoogsteen sites in supramolecular architectures of some coordination metal complexes of N(6)-benzyl/furfuryl adenine. - Chemistry Central journal
Most of the benzyladenine and furfuryladenine derivatives inhibit tumor/cancer cell growth; their toxicity is lesser than the compounds used for the treatment of cancer now-a-days. Many cytokinin derivatives are tested for anticancer activity.A series of transition metal complexes containing N(6)-benzyl/furfuryl aminopurines of formula [Mn(FAH)2(H2O)(Cl3)]2.Cl2 (1), [Co(FAH)2(H2O)(Cl3)]2.Cl2 (2), [Co(FAH)2(Cl4)]2 .[Co(FAH)2(H3O)(Cl3)].Cl2 (3), [Ni(FAH)2(H2O)(Cl3)]2.Cl2. (H2O) (4), [Zn(BAH)Br3] (5) and [Cd2(BAH)2(μ-Br)4Br2]n (6) (where BAH and FAH benzyladeninium and furfuryladeninium cations respectively) have been synthesized and characterized. Crystal structures of (1-4) have similar distorted octahedral coordination geometry, while (5) and (6) have distorted tetrahedral geometry and octahedral geometries respectively. In (1-4) two halide ions and two cytokinin cations (BAH(+)/FAH(+)) are laterally coordinated to the metal ion. A water molecule and a halide ion are axially coordinated. But the coordination sphere of (5) consists of N7 coordinated benzyladeninium ion and three halide ions. The complex (6) is a coordination polymer bridged by bromide anions. A common notable feature in (1-4) is the presence of one or more lattice chloride anions. They help in a chain formation by N-H…Cl halide involving hydrogen bonding interactions in between the Hoogsteen site hydrogen.The observed crystal structures emphasize the role of the halide ions in developing the supramolecular architectures by halide involving hydrogen bonding interactions. Also most of the reported cobalt cytokinin complexes possess tetrahedral coordination geometry, but some cobalt complexes have distorted octahedral coordination geometry, which are discussed and compared. Targeting SUMOylation cascade for diabetes management. - Current drug targets
Post-translational modifications (PTMs) play important roles in regulating protein stability, trafficking, folding conformation, and functional activity. Small ubiquitin-like modifier (SUMO) protein mediates a distinct type of PTM called SUMOylation in which the SUMO protein is covalently ligated to the target protein and modifies its activities through a series of enzymatically-catalyzed reactions. SUMOylation regulates many cellular processes like transcription, the maintenance of the ion gradient across the cell membrane, stress response, autoimmunity, etc. Several target proteins of SUMOylation are involved in the biological pathways related to various human diseases, including cardiovascular diseases, diabetes, cancer, and neurodegenerative disorders. This review focuses on the SUMOylation process, regulatory roles of SUMOylation in diabetes, and prospects of developing novel anti-diabetic drugs targeting the SUMOylation process.
Multiplexed analysis of protein-ligand interactions by fluorescence anisotropy in a microfluidic platform. - Analytical chemistry
Homogeneous assay platforms for measuring protein-ligand interactions are highly valued due to their potential for high-throughput screening. However, the implementation of these multiplexed assays in conventional microplate formats is considerably expensive due to the large amounts of reagents required and the need for automation. We implemented a homogeneous fluorescence anisotropy-based binding assay in an automated microfluidic chip to simultaneously interrogate >2300 pairwise interactions. We demonstrated the utility of this platform in determining the binding affinities between chromatin-regulatory proteins and different post-translationally modified histone peptides. The microfluidic chip assay produces comparable results to conventional microtiter plate assays, yet requires 2 orders of magnitude less sample and an order of magnitude fewer pipetting steps. This approach enables one to use small samples for medium-scale screening and could ease the bottleneck of large-scale protein purification.
Syntheses and characterization of two novel tetranuclear lead(II) clusters self-assembled by hydrogen bonded interactions. - Chemistry Central journal
The usage of polynuclear metal clusters as secondary building units (SBU's) in designing of metal organic frameworks (MOF's) is a field of current interest. These metal clusters have attracted a great deal of attention not only due to their interesting structural topologies but also due to promising physical and chemical properties. In this regard various d,f block (transition and lanthanide) metal clusters have been widely investigated so far. Less attention is paid to construction of heavy p-block Pb(II) clusters.Two mixed ligand Pb(II) clusters have been synthesized with bipy(2,2'-Bipyridine), phen(1,10-Phenanthroline), quin (8-Hydroxy quinolinate) and 5-tpc (5-chloro thiophene 2-carboxylate). They have been characterized by elemental analysis, IR, TGA and X-ray crystallography. X-ray diffraction analysis reveals that the complexes [Pb4(quin)4(bipy)2(5-tpc)4] (1) and [Pb4(quin) 4(phen) 2(5-tpc)4] (2) are tetranuclear. The complexes show a slight variation in unit cell parameters, due to the replacement of bipy and the phen ligands. Both complexes contain two types of Pb(II) ions which differ in the coordination geometry around the Pb(II) ion.In both complexes the four lead ions Pb1,Pb2, Pb1(i) and Pb2(i) lie on the same plane bridged by the 5-tpc anions. Pb1 and Pb2 of both complexes contain a 5-tpc and quin coordinated in a bidentate chelating bridging fashion. In addition the Pb2 and Pb2(i) ions alone contain a bipy and phen in a bidentate chelating fashion in (1) and (2) respectively. An additional notable feature in both of these complexes are the bridging ability of the quin oxygen which forms a network of coordination bonds in between the four Pb(II) ions. In both complexes the individual units are self-assembled by C-H---Cl/C-H---S hydrogen bonding interactions to generate 2-D aggregates.
Design of co-crystals/salts of some Nitrogenous bases and some derivatives of thiophene carboxylic acids through a combination of hydrogen and halogen bonds. - Chemistry Central journal
The utility of N-heterocyclic bases to obtain molecular complexes with carboxylic acids is well studied. Depending on the solid state interaction between the N-heterocyclic base and a carboxylic acid a variety of neutral or ionic synthons are observed. Meanwhile, pyridines and pyrimidines have been frequently chosen in the area of crystal engineering for their multipurpose functionality. HT (hetero trimers) and LHT (linear heterotetramers) are the well known synthons that are formed in the presence of pyrimidines and carboxylic acids.Fourteen crystals involving various substituted thiophene carboxylic acid derivatives and nitrogenous bases were prepared and characterized by using single crystal X-ray diffraction. The 14 crystals can further be divided into two groups [1a-7a], [8b-14b] based on the nature of the nitrogenous base. Carboxylic acid to pyridine proton transfer has occurred in 3 compounds of each group. In addition to the commonly occurring hydrogen bond based pyridine/carboxylic acid and pyrimidine/carboxylic acid synthons which is the reason for assembly of primary motifs, various other interactions like Cl…Cl, Cl…O, C-H…Cl, C-H…S add additional support in organizing these supermolecules into extended architectures. It is also interesting to note that in all the compounds π-π stacking occurs between the pyrimidine-pyrimidine or pyridine-pyridine or acid-acid moieties rather than acid-pyrimidine/pyridine.In all the compounds (1a-14b) either neutral O-H…Npyridyl/pyrimidine or charge-assisted Npyridinium-H…Ocarboxylate hydrogen bonds are present. The HT (hetero trimers) and LHT (linear heterotetramers) are dominant in the crystal structures of the adducts containing N-heterocyclic bases with two proton acceptors (1a-7a). Similar type supramolecular ladders are observed in 5TPC44BIPY (8b), TPC44BIPY (9b), TPC44TMBP (11b). Among the seven compounds [8b-14b] the extended ligands are linear in all except for the TMBP (10b, 11b, 12b). The structure of each compound depends on the dihedral angle between the carboxyl group and the nitrogenous base. All these compounds indicate three main synthons that regularly occur, namely linear heterodimer (HD), heterotrimer (HT) and heterotetramer (LHT).
Toxicity effect of silver nanoparticles in brine shrimp Artemia. - TheScientificWorldJournal
The present study revealed the toxic effect of silver nanoparticles (AgNPs) in Artemia nauplii and evaluated the mortality rate, hatching percentage, and genotoxic effect in Artemia nauplii/cysts. The AgNPs were commercially purchased and characterized using field emission scanning electron microscope with energy dispersive X-ray spectroscopy. Nanoparticles were spherical in nature and with size range of 30-40 nm. Artemia cysts were collected from salt pan, processed, and hatched in sea water. Artemia nauplii (II instar) were treated using silver nanoparticles of various nanomolar concentrations and LC50 value (10 nM) and mortality rate (24 and 48 hours) was evaluated. Hatching percentage of decapsulated cysts treated with AgNPs was examined. Aggregation of AgNPs in the gut region of nauplii was studied using phase contrast microscope and apoptotic cells in nauplii stained with acridine orange were observed using fluorescence microscope. DNA damage of single cell of nauplii was determined by comet assay. This study showed that as the concentration of AgNPs increased, the mortality rate, aggregation in gut region, apoptotic cells, and DNA damage increased in nauplii, whereas the percentage of hatching in Artemia cysts decreased. Thus this study revealed that the nanomolar concentrations of AgNPs have toxic effect on both Artemia nauplii and cysts.
